View All» Immunogen: Linear peptide corresponding to human YB-1. View All» Background Information: Y-box-binding protein 1 (YB-1) is a member of the highly conserved Y-box protein family. This multifunctional protein binds single stranded DNA and RNA and functions as a regulator of transcription, RNA metabolism, and protein synthesis. YB-1 promotes the separation of DNA strands containing mismatches or modifications by cisplatin. In vitro studies have also shown that YB-1 has endonucleolytic activity resulting in the introduction of nicks or breaks into double-stranded DNA. YB1 may play a role in DNA repair. YB1 binds to Y-box sequences present in multiple gene promoters to positively or negatively regulate transcription. YB1 activates genes associated with proliferation and cancer, such as cyclin A, cyclin B1, and matrix metalloproteinase-2 and represses genes associated with cell death, such as the FAS cell death-associated receptor and the p53 tumor suppressor gene.In RNA, YB-1 binds to splice sites in pre-mRNA and plays a role in determining splice site selection. The interaction of this protein with RNA in the cytoplasm stabilizes mRNA and appears to contribute to the regulation of translation by modulating the interaction between the mRNA and eukaryotic initiation factors.
