Preparation and Storage

The antibody was purified from tissue culture supernatant by affinity chromatography. The antibody solution should be stored undiluted at 4°C. Since this preparation contains no preservative, it should be handled under aseptic conditions.

Usage

This antibody has been tested by LAL assay for endotoxin level and by immunofluorescent staining ( 1 mg/million cells) with flow cytometric analysis to assure specificity and reactivity. Since this antigen is expressed at low density on lymphoid cell surfaces, it may be desirable to amplify staining by using a bright second-step reagent, such as PE-conjugated donkey anti-rat Ig (Cat. no. 559970) polyclonal antibody. If a three-step method is employed, we recommend the use of Mouse Fc Block™ (Cat. no. 553141/553142) to reduce non-specific staining. In this case, a second-step antibody which does not cross-react with 2.4G2 mAb (rat IgG_2b, k) must be used. We recommend biotinylated anti-rat IgG_2a (RG7/1.30, Cat. no. 553894) followed by Streptavidin-PE (Cat. no. 554061). Other reported applications include blocking of functional assays.¹¹ We do not recommend the SB/14 mAb for western blot analysis, immunoprecipitation, or immunohistochemical staining. Since applications vary, each investigator must determine dilutions appropriate for individual use.
