白细胞介素-2(IL-2)是一种最早发现的具有广泛生物活性的细胞因子，它必须和IL-2受体结合才能发挥生物学效应。白细胞介素-2受体(IL-2R)是由3个糖蛋白亚基组成的，分别为 α，β，γ链，分子量为55kD，75kD和64kD，故又称p55，p75和p64。单个亚基不能传递IL-2信号，只有2或3个亚基组成复合体方能发挥受体功能。大量交叉实验表明，IL―2的种属特异性是有一定规律的，一般呈下行性，即进化程度高的动物的IL-2可以作用于进化程度较低的动物。如人和猿的IL-2。几乎可以作用于所有哺乳动物的细胞；但其它哺乳动物的IL-2很少可以作用于人。由于IL-2的生物学作用是以刺激T细胞增生为主，故又称T细胞生长因子，除此之外还有其它重要功能。

人白介素-2重组蛋白是在大肠杆菌中重组表达的，是一条单链、非糖基化的突变蛋白，包含有134个氨基酸，其中在126位的Cys氨基酸被替换成了Ser，分子量为15517Da。该人白介素-2重组蛋白最终由标准的色谱技术纯化出。

溶解：提供的人白介素-2(IL 2)产品为过滤灭菌的白色冻干粉末；该冻干粉末是从含1.1mg/ml人白介素-2(IL 2)的 0.17mg磷酸二氢钠和0.89mg磷酸氢二钠缓冲液（pH 7.5）中经进一步透析并冻干产生的。我们建议加无菌的20mM AcOH以配制工作储存液到100ug/ml，后继实验过程中可根据实验要求进一步稀释。

保存：人白介素-2(IL 2)冻干粉末在室温可保存3周，或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天；如果想延长保存时间，建议分装（加载体蛋白如0.1% HSA或 BSA）后置于-18℃冻存。请避免反复冻融。

Introduction

IL2 is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.

Description

Interleukin-2 Human Recombinant produced in E.Coli is a single, non-glycosylated mutein (variant form) of human IL-2 polypeptide chain containing 134 amino acids and having a molecular mass of 15517 Dalton.

Our Interleukin -2 has an Ser substitute for Cysteine at position 126.

The IL-2 is purified by proprietary chromatographic techniques.

Physical Appearance

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation

The protein (1.1mg/ml) was lyophilized after extensive dialysis against 0.17mg sodium monobasic & 0.89mg dibasic sodium phosphate buffer to a pH=7.5.

Solubility

It is recommended to reconstitute the lyophilized Interleukin 2 in sterile 20mM AcOH not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability （稳定性）

Lyophilized Interleukin-2 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IL2 should be stored at 4°C between 2-7 days and for future use below -18°C.

For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).

Please prevent freeze-thaw cycles.

Greater than 98.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Analysis by SDS-PAGE.

Amino acid sequence （氨基酸序列）

The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Pro-Thr-Ser.

Biological Activity The ED50 as determined by the dose-dependant stimulation of murine CTLL-2 cells is < 0.0645 ng/ml, corresponding to a Specific Activity of 15,500,000IU/mg.

Protein content Protein quantitation was carried out by two independent methods

1. UV spectroscopy at 280 nm using the absorbency value of 0.614 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).

2. Analysis by RP-HPLC, using a calibrated solution of IL-2 as a Reference Standard.
