It is expressed on postsynaptic neurons and initiates excitatory presynapse maturation through calcium-dependent binding to a select isoform of beta-neurexin. The 848 amino acid (aa) human NLGN3 contains a 38 aa signal sequence and a 670 aa extracellular domain that shares 99.7% aa identity with mouse and rat NLGN3. Alternate splice forms of human NLGN3 encode 828, 808 and 711 aa lacking aa 153-172, 153-192, and 1-118 (plus 153-172), respectively. Certain NLGN3 mutations are associated with rare cases of autism.
