    View All» Application Notes: Immunofluorescence and immunoperoxidase staining of formalin-fixed and paraffin-embedded sections of anterior pituitary glands. Cryostat sections of the same tissues can also be used.Optimal working dilutions must be determined by the end user.IMMUNOPEROXIDASE STAINING:1. Incubate the section with antiserum diluted 1:1000 - 1:2,000 in PBS at 37°C for 30 minutes.2. Wash for 15 minutes with PBS at room temperature and treat sequentially with Goat anti-Rabbit IgG and PAP complex at 37°C. Wash excess reagent from the section as described above.3. Treat the section with a mixture of 3,3'-diaminobenzidine HCl (0.02%) in hydrogen peroxide (0.004%). For preservation, the sections may be dehydrated, cleared and mounted.IMMUNOFLUORESCENCE STAINING:1. Same as Step 1 above.2. Incubate the section with antiserum diluted 1:20-1:40 in PBS at 37°C for 30 minutes.3. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed in the pituitary but not in placental tissue as is the case for the other four genes in the growth hormone locus.
