DKK-1是重组人Dickkopf相关蛋白1的简称,是DKK蛋白家族(DKK1-4)的一员。DKK最初被认为是非洲爪蟾蜍的前期成形细胞。通过机理研究发现DKK-1通过与共同受体LRP5/6一起形成抑制复合物,可以有效抑制Wnt/β-catenin信号通路。研究人员发现敲除DKK-1基因小鼠的脑部无法形成,从而证实Wnt/β-catenin信号通路的抑制在脊椎动物的后模式与前模式发展中必不可少。
StemRD公司生产的人重组DKK-1蛋白来源于人的293细胞,是含有235个氨基酸片段,分子量为3740kDa的糖蛋白。通过诸如HPLC等一系列色谱分析方法对其进行纯化。

DKK-1, Dickkopf-related protein-1, is a member of the DKK protein family which includes DKK-1 through DDK-4. DKK was originally identified as a head-forming molecule in Xenopus. Mechanistic studies showed that DKK-1 inhibits the Wnt /β-catenin signaling pathway by forming inhibitory complexes with co-receptor LRP5/6. Inhibition of Wnt/β-catenin signaling is essential for posterior patterning and anterior development in vertebrates, supported by the discovery that DKK-1 knockout mice lack head formation.
StemRD expressed recombinant human DKK-1 in human 293 cells as a 37-40 kDa glycoprotein containing 235 amino-acid residues. It is purified by aseries of chromatography including HPLC.
• Purity: Greater than 90% as determined by SDS-PAGE analysis
• Biological Activity: The activity was determined by using a TCF reporter gene assay in cultured human cells. The IC50 ranges from 100 - 500 ng/ml in the inhibition of 100 ng/mL WNT-3a activity. Activity in other assays should be determined by each individual setting.
