View All» Background Information: RING Finger protein 8 (RNF8) is DNA binding protein belonging to the RING (Really Interesting New Gene) finger group of proteins. This family is characterized by a RING finger domain containing a Cys3HisCys4 amino acid motif capable of binding two zinc cations. RNF8 contains two domains of interest, the characteristic RING finger motif in the C-terminus and a FHA domain within the N-terminus, and is a critical E3 ubiquitin-protein ligase involved in the assembly of repair proteins to damaged DNA sites. After DNA double-strand breaks, RNF8 is recruited by ATM-phosphorylated MDC1 to the damage sites, where it functions to mediate H2A and H2AX ubiquitination, which in turn promotes TPR53BPI formation and the ionizing radiation-induced foci (IRIF) of BRCA1.
