Temperature-sensitive mutations of this protein have been associated with defects in sister chromatid cohesion, chromosome condensation, and chromosome segregation. Its expression is cell cycle regulated and peaks in S phase. Expression of MDC1 is required for the S- and G2/M-checkpoint-mediated cell cycle arrest in response to DNA damage. MDC1 has been shown to interact with H2A.X near sites of DNA double-strand breaks and recruit ATM kinase to the damaged region of DNA.
