Two distinct but chemically related IGFs have been purified from human plasma: IGF-I (Somatomedin C) and IGF-II. They are single chain polypeptides with molecular weights of 7,640 and 7,471, respectively. Types I and II IGF receptors differ in their structure and in their peptide binding specificity. Type I IGF receptors are structurally homologous to insulin receptors and contain alpha-subunits (MW = 130,000) that bind peptide, and beta-subunits (MW = 90,000), which have intrinsic tyrosine kinase activity. The protein-tyrosine kinase activity of the insulin receptor and, presumably the IGF-I receptor, is required for insulin to stimulate all of its biological responses in cells.
