Atg12 is the human homolog of a yeast protein involved in autophagy (1). It is essential for Atg12 conjugation, and the Atg12-Atg5 conjugate is necessary to form autophagic vesicles (2). The E3-like activity of the Atg12-Atg5 conjugate potentiates the formation of Atg8-PE; making the autophagic pathway analogous to ubiquitin-like pathways where an E3 conjugation ligase is required (3).
