Human type A erthrocytes (washed and suspended at a concentration of 2% v/v) are prepared, and 50μL are added to the virion-antibody mixtures, and incubated overnight at 4C; The HA inhibition (HAI) titer is defined as the lowest concentration of antibody capable of inhibiting HA. For 9BG5 this titer is typically >3 μg/mL {Nason, et al, 2001}. Dilute with buffer pH 7.5 - 8.0 to desired working volumes.
