CENP-E, along with cytoplasmic dyenin, is one of two main microtubule motors known to localize at kinetochores. During mitosis, CENP-E has an active role in the migration of chromosomes toward the metaphase plate, and is directly responsible for microtubule capture and stabilization by kinetochores. Reduction in CENP-E expression results in the high occurrence of whole chromosome missegregation, and its removal or inhibition leads to the presence of unattached chromosomes near the spindle pole as well as a significant reduction in the number of microtubules captured by kinetochores.
