PDX-1 is a critical factor in the processes of development and differentiation of the pancreas, specifically through the designation of early pancreatic epithelium, allowing it to proliferate, branch and ultimately differentiate. It has been shown to stimulate gene transcription for insulin, islet amyloid polypeptide, glucokinase, glucose transporter type 2, and somatostatin. PDX-1 also serves to regulate beta cells past development and into the adult stage. Defects in PDX-1 are the cause of several pancreatic diseases including; pancreatic agenesis, maturity-onset diabetes of the young type 4, and non-insulin-dependent diabetes mellitus. Up-regulation of PDX-1 in adult tissue has been associated with pancreatitis and pancreatic cancer. PDX-1 is expressed in the duodenum and pancreas tissues.
