SUMO (small ubiquitin-like modifier) functions in a manner similar to ubiquitin in that it is bound to target proteins as part of a post-translational modification system. Sentrin is a ubiqutin-like protein that interacts with the death domains of Fas, tumor necrosis factor receptor 11, the tumor suppressor PML, Rad 51 and Rad52, which are involved in repairing dsDNA breaks, and GAP1. This interaction requires the activation of Sentrin’s conserved C-terminal Gly-Gly residues, which is very similar to the ubiquitin pathway. Sentrin can be conjugated to other proteins in a manner analogous to protein ubiquitination. Sentrinized proteins are localized in the nucleus, whereas the sentrin 18 kDa monomer resides mostly in the cytosol. The presence of sentrin-modified p90, appears to be a prerequisite for sentrin modification of nuclear proteins to occur. Over-expression of sentrin in mammalian cells appears to protect cells against both anti-Fas and TNF-induced cell death. Sentrinization represents a novel pathway for nuclear protein modification, which is distinct from ubiquitination. 规格
