It is suitable for use in analysis of protein structure, studying protein-protein interactions, and cell biology research applications. It can also be used as an immunogen, or as a protein standard. This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Thr 211) of human TNFRSF1A (NP_001056.1) fused to a polyhistidine tag at the C-terminus.Activity: 1. Measured by its binding ability in a functional ELISA: a. Immobilized human TNFa at 10 µg/ml(100 µl/well) can bind biotinylated human TNFRSF1A-his. The ED₅₀ of biotinylated human TNFRSF1A-his is 0.04 µg/ml. b. Immobilized mouse TNFa at 10 µg/ml(100 µl/well) can bind biotinylated human TNFRSF1A-his. The ED₅₀ of biotinylated human TNFRSF1A-his is 0.28 µg/ml. 2. Measured by its ability to inhibit the TNF mediated cytotoxicity in the L929 mouse fibrosarcoma cells in the presence of the metabolic inhibitor actinomycin D. The ED₅₀ for this effect is typically 0.04-0.2 µg/mlin the presence of 0.25 ng/mlrecombinant human TNF.Formulation: Lyophilized in PBS, pH 7.4, 5% Mannitol, 5% Trehalose, and 0.02% Tween 80.Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.
